榆栀止血颗粒通过HIF-1α/VEGF通路缓解肾阴虚型异常子宫出血

Abstract
Objective:To investigate the effect of Yuzhi Zhixue Granule(榆栀止血颗粒,YZ)on uterus of abnormal uterine bleeding(AUB)rats with Shenyinxu(肾阴虚)syndrome and explore its mechanism.Methods:A total of 36 female SD rats were randomly divided into a control group,a model group,and a 4.5 g/kg YZ group.The model of AUB with Shenyinxu syndrome was established,and rats in the YZ group were orally administered YZ solution for 15 consecutive days.After the end of administration,the rats were sacrificed at the late stage of estrus.In each group,4 rats were randomly selected,and barium sulfate was used in vascular perfusion to observe and quantify uterine angioarchitecture by micro-computed tomography(micro-CT).The blood and uteri of the remaining rats were collected.The levels of serum follicle-stimulating hormone(FSH),anti-Müllerian hormone(AMH),estradiol(E2),and progesterone(P)were measured by enzyme-linked immunosorbent as-say(ELISA).The pathological changes of uterine were measured by hematoxylin-eosin(HE)staining.The protein expression of hypoxia-in-ducible factor-1α(HIF-1α)and vascular endothelial growth factor(VEGF)in uterine tissue was detected by Western blot(WB).Results:Compared with the control group,the model group showed the vaginal bleeding symptom,an significantly increased serum FSH level(P<0.01),significantly decreased levels of AMH,E2,P and proportion of vascular volume in uterus(P<0.05 or P<0.01),an elevated propor-tion of microvascular volume(P<0.05)but significantly decreased number of tertiary and quaternary vessels(P<0.01 or P<0.05),and down-regulated protein expression of HIF-1α and VEGF in uterine tissue(P<0.01).Compared with the model group,the 4.5 g/kg YZ group showed no obvious symptom of vaginal bleeding,a significantly decreased serum FSH level(P<0.05),significantly increased levels of AMH,E2,and P,proportion of vascular volume in uterus,number of quaternary vessels,and protein expression of HIF-1α and VEGF(P<0.05 or P<0.01).Conclusion:YZ can improve uterine bleeding symptoms and hormone levels in AUB rats with Shenyinxu syndrome,which may be re-lated to increasing the number of microvessels,decreasing microvascular dilatation,and regulating uterine angioarchitecture through HIF-1α/VEGF signaling pathway.关键词
