参七糖络方调控PI3K/AKT信号通路缓解2型糖尿病
Alleviating Effect of Shenqi Tangluo Formula on Type 2 Diabetic Rats Based on PI3K/Akt Signaling Pathway

Abstract
Objective:To explore the mechanism of Shenqi Tangluo(参七糖络)Formula in improving glucose and lipid metabolism in type 2 dia-betes mellitus based on the phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt)pathway.Methods:Eighty SD rats were randomly divided into a normal group and a model group.Rats in the normal group were fed with normal diet,while the rats in the model group received a high-sugar and high-fat diet combined with intraperitoneal injection of low-dose streptozotocin to replicate the rat model of type 2 diabetes mellitus.The successfully modeled rats were divided into a model group,a metformin hydrochloride(180 mg/kg)group,as well as 13.4,26.8,and 53.6 g/kg Shenqi Tangluo Formula groups,with 12 rats in each group.All groups were constantly administered corresponding medi-cines or saline water by gavage for 28 days.After administration,the following aspects were determined:fasting blood glucose(FBG),serum fasting insulin(FINS),total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),and high density lipoprotein cholesterol(HDL-C)by enzyme-linked immunosorbent assay(ELISA),homeostasis model assessment of insulin resistance(HOMA-IR),pathological changes in rat liver tissue under the electron microscope,the PI3K,p-PI3K,AKT,p-AKT,FOXO1,p-FOXO1,GSK-3β,and p-GSK-3β protein expression levels in liver tissue by Western blot,as well as the FoxO1,and GSK-3β expressions in liver tissue by immunohis-tochemistry.Results:Compared with the normal group,the FBG of the model group was significantly increased(P<0.01),while FINS was significantly decreased.Additionally,the lipid metabolism was abnormal,with significantly increased serum TC,TG,and LDL-C levels(P<0.05),as well as significantly decreased HDL-C level(P<0.05).Liver cell steatosis and notable pathological changes were observed.The PI3K,Akt,p-PI3K/PI3K,p-Akt/Akt,GSK-3β,and p-GSK-3β/GSK-3β protein expression levels in liver tissue were significantly decreased(P<0.05),while the FoxO1 and p-FoxO1/FoxO1 protein expression levels were significantly increased(P<0.05).Compared with the model group,the glucose and lipid metabolism of rats in Shenqi Tangluo Formula groups of each dose were significantly improved(P<0.05).The pathological state of liver cells was improved,the PI3 K,Akt,p-PI3 K/PI3 K,p-Akt/Akt,GSK-3β,and p-GSK-3β/GSK-3β protein expression levels in liver tissue were significantly increased(P<0.05),while the FoxO1,p-GSK-3β,and p-FoxO1/FoxO1 protein expression levels were significantly decreased(P<0.05),and the Foxo1,G6pase,and Pepck mRNA expression levels in liver tissue were significantly decreased(P<0.05).Conclusion:Shenqi Tangluo Formula can effectively improve glucose and lipid metabolism in T2DM rats and the pathological con-dition of liver tissue.The mechanism may be related to the PI3K/Akt pathway.关键词
