摘要

Abstract

Objective:To explore the mechanism of Yangxin Yinxie(养心银屑)Prescription in treating psoriasis based on the transforming growth factor(TGF)-β/Smad signaling axis.Methods:The psoriasis model was established by applying imiquimod on the back of mice.Mice were randomized into control,model,Yangxin Yinxie Prescription(17,34,and 68 g/kg),and methotrexate(1 mg/kg)groups,with 6 mice in each group.While modeling,mice were administrated with corresponding drugs or normal saline,once a day for 7 consecutive days.The lesion area was observed and the psoriasis area severity index(PASI)was scored.The pathological changes in skin lesions were observed by hematoxylin-eosin staining.The levels of interleukin(IL)-17,IL-6,tumor necrosis factor(TNF)-α,and interferon(IFN)-γ in the serum were measured by ELISA.The expression of Ki-67 was observed by immunohistochemical staining.Flow cytometry was employed to detect the proportions of helper T cell 17(Th17+)and regulatory T cell(Treg+)in the peripheral blood.The mRNA levels of RORγt and Foxp3 in skin lesions were determined by qRT-PCR.Western blotting was employed to determine the protein levels of TGF-β1,TβRⅡ,Smad3,and Smad4 in skin le-sions.Results:Compared with the control group,the model group showed psoriasis characteristic pathological changes such as thickened epi-dermis,hyperkeratosis,parakeratosis,and inflammatory cell infiltration,increases in the PASI score of skin lesions(P<0.01)and the propor-tion of Th17+cells in the peripheral blood,a decrease in the proportion of Treg+cells,and elevated serum levels of IL-17,IL-6,TNF-α,and IFN-γ(P<0.01).In addition,the modeling down-regulated the mRNA level of Foxp3 while up-regulating the mRNA level of RORγt in skin lesions,and it down-regulated the protein levels of TGF-β1,TβRⅡ,Smad3,and Smad4(P<0.01).Compared with the model group,Yangxin Yinxie Prescription(17,34,and 68 g/kg)alleviated the epidermal thickening,hyperkeratosis,parakeratosis,and spinous layer hypertrophy,decreased the proportion of Th17+cells and increased the proportion of Treg+cells in the peripheral blood,and lowered the serum levels of IL-17,IL-6,TNF-α,and IFN-γ(P<0.01).Yangxin Yinxie Prescription at 68 g/kg decreased the PASI score of skin lesions(P<0.01),up-reg-ulated the mRNA level of Foxp3,down-regulated the mRNA level of RORγt,and up-regulated the protein levels of TGF-β1,TβRⅡ,Smad3,and Smad4(P<0.01).Conclusion:Yangxin Yinxie Prescription may treat psoriasis by regulating the Th17/Treg cell balance mediated by the TGF-β/Smad signaling axis.
