Effect of ACANTHOPANACIS SENTICOSI RADIX ET RHIZOMA SEU CAULIS Extract on Apoptosis in α-Synuclein Overexpressing SHSY5Y Cells

Abstract
Objective:To investigate the effect of ACANTHOPANACIS SENTICOSI RADIX ET RHIZOMA SEU CAULIS extract on apoptosis in a lentivirus-transfected human neuroblastoma cell PD model.Methods:SHSY5Y cells were transfected with lentivirus to construct a PD cell model overexpressing α-synuclein.The effects of Acanthopanax senticosus extract on SHSY5Y cells were observed.The experiment was divid-ed into four groups:blank control,unloaded lentivirus control,model control,and ACANTHOPANACIS SENTICOSI RADIX ET RHIZOMA SEU CAULIS 6 mg/mL treatment group.The optimal dosage was determined using the CCK-8 assay.Intracellular reactive oxygen species(ROS)levels were measured using dihydroethidium staining,cell viability was assessed with a cell viability assay kit,calcium ion concentra-tion was evaluated using a calcium colorimetric assay kit,and mitochondrial membrane potential was measured with the TMRE mitochondrial membrane potential assay kit.Protein expression levels of Bcl-2,Bax,caspase-3,PI3K,p-PI3K,AKT,p-AKT,MCU,and Cytc were assessed by Western blotting.Results:Compared with the blank control and empty lentivirus groups,the ROS level of cells in the model control group increased,ATP and mitochondrial membrane potential(MMP)decreased,calcium ion content increased(P<0.01),the expression of phos-phorylated PI3K,AKT,BCL2,and mitochondrial Cytc proteins were downregulated,and the expression of BAX,Caspase3,MCU,and cy-tosolic Cytc proteins were upregulated(P<0.01).Compared with the model control group,the ACANTHOPANACIS SENTICOSI RADIX ET RHIZOMA SEU CAULIS treatment group showed a significant reduction in ROS levels(P<0.01)and a significant increase in cell viability(P<0.01).Calcium ion concentration decreased significantly(P<0.01),and mitochondrial membrane potential increased significantly(P<0.01).Expression levels of Bax,caspase-3,MCU,and cytoplasmic Cytc were significantly reduced(P<0.01),while p-PI3K/PI3K,p-AKT/AKT,Bcl-2,and mitochondrial Cytc protein expression increased significantly(P<0.01).Conclusion:ACANTHOPANACIS SENTICOSI RA-DIX ET RHIZOMA SEU CAULIS extract can reduce apoptosis in human neuroblastoma cells,potentially exhibiting neuroprotective effects.关键词
