摘要

Abstract

Objective:To explore the mechanism by which Raphani Semen ameliorates liver injury in high-normal blood pressure(PHT)rats with phlegm-dampness stagnation syndrome through its effects on FGF15 and FGFR4 expression in the liver and FXR and FGF15 expression in the ileum.Methods:PHT rats with phlegm-dampness stagnation syndrome were used as experimental models,fed a high-fat diet,and injected intraperitoneally with L-NNA to establish the model.After modeling,rats were randomly divided into the model group(n=15),Raphani Semen group(n=15),and silymarin group(n=15).The Raphani Semen group(2.5 g/kg)and silymarin group(20 mg/kg)were treated with corresponding interventions once daily.After six weeks,liver tissue morphology was observed,and protein expression levels of FXR and FGF15 in the ileum and FGF15 and FGFR4 in the liver were detected.Results:The model group exhibited a large number of lipid droplets and vacuoles in the liver,while the Raphani Semen and silymarin groups showed significant reductions in fat vacuoles.FGF15 and FGFR4 expression in the liver and FXR and FGF15 expression in the ileum were lower in the model group than in the control group(P<0.05).The Raphani Semen group exhibited higher expression of these proteins compared to the model group(P<0.05).Conclusion:Raphani Semen exerts significant protective effects on liver injury in PHT rats with phlegm-dampness stagnation syndrome.The mechanism involves FXR activation,which induces FGF15 expression,increases FGF15 levels in the enterohepatic circulation,and activates the FGFR4 signaling pathway to exert protective effects.
