Abstract

Objective:To explore the anti-inflammatory mechanism and the effects on cell migration and invasion of Zuojin pill in Helicobacter pylori(Hp)-associated precancerous gastric lesions(PLGC)based on miR-107.Methods:An Hp infected GES-1 cell model was established and divided into the model group,negative control group,and low-dose(10%),medium-dose(20%),and high-dose(40%)Zuojin pill groups.Cell migration,invasion capabilities,and miR-107 gene expression levels were measured.A total of 70 rats were used,with 10 rats as the blank group(given physiological saline 3 mL/kg via gavage),and the remaining rats underwent multifactorial modeling to establish the PLGC model.Among the 41 successfully modeled rats,14 were assigned to the model group(given physiological saline 3 mL/kg via gavage),14 to the Weimeisu group(Vitacoenzyme Suspension,2 mL/kg),and 13 to the Zuojin pill group(Zuojin pill decoction,3 mL/kg).Treatments were administered once daily for 12 weeks.Western blotting was performed to detect the relative protein expression levels of IL-6,STAT3,p-STAT3,JAK2,and p-JAK2.Results:Compared with the negative control group,the model group showed significantly increased cell migration distance and miR-107 gene expression levels(P<0.05).Compared with the model group,the low,medium,and high-dose Zuojin pill groups exhibited significantly reduced invaded cell counts,migration distances,and miR-107 gene expression levels(P<0.05).These reductions were dose-dependent,with the high-dose group showing the most pronounced effects(P<0.05).Compared with the blank group,the model group rats exhibited significantly elevated levels of IL-6,p-STAT3/STAT3,and p-JAK2/JAK2 protein expression(P<0.05).Compared with the model group,the Weimeisu group and Zuojin pill group showed reduced IL-6,p-STAT3/STAT3,and p-JAK2/JAK2 protein expression levels(P<0.05).Compared with the Weimeisu group,the Zuojin pill group exhibited further reductions in these protein expression levels(P<0.05).Conclusion:Zuojin pill demonstrates effective anti-inflammatory and anticancer effects in the Hp infection-associated PLGC rat model.The mechanism may involve inhibiting miR-107 and suppressing the activation of the IL-6/JAK2/STAT3 pathway,thereby reducing cell migration and invasion.
