摘要

Abstract

Objective:To investigate the therapeutic effects and mechanism of Modified Maxing Yigan Decoction(MMYD)on post-infectious cough(PIC)through PI3K/Akt/NF-κB pathway.Methods:Forty-eight female Balb/c mice were randomly divided into normal group,model group,low-dose MMYD group(50 mg/kg),medium-dose MMYD group(100 mg/kg),high-dose MMYD group(200 mg/kg),and positive control group(Asmeton capsules,20 mg/kg)with 8 mice per group.Except the normal group,other groups were exposed to cigarette smoke in a chamber for 30 min daily for 10 days.From day 11 to 19,PIC models were established by intranasal instillation of 0.4 mg/mL LPS(10 μL/10 g)to simulate Gram-negative bacterial infection.Treatments were administered from day 11 for 9 days.Serum and lung tissues were collected for ELISA analysis of inflammatory factors,HE staining for pathological evaluation,and Western blot/immunohistochemistry/immunofluorescence for PI3K/Akt/NF-κB pathway protein detection.Results:ELISA showed serum IL-1β,IL-6 and TNF-α levels in model group were significantly higher than normal group(P<0.01),while all treatment groups showed significant reduction(P<0.05).Serum IL-10 was significantly lower in model group(P<0.001)but increased after treatment.HE staining revealed significant inflammatory cell infiltration,epithelial metaplasia and airway wall thickening in model group,which were alleviated in MMYD groups.Immunohistochemistry showed elevated mTOR expression in model group(reduced in treatment groups).Immunofluorescence demonstrated increased p-PI3K and p-Akt in model group,with significant reduction in medium/high-dose and positive groups.Western blot showed elevated IκBα,p-IκBα,p65 and p-p65 in model group,all decreased after treatment.Conclusion:MMYD exerts anti-inflammatory and antitussive effects by inhibiting PI3K/Akt/NF-κB pathway activation,reducing inflammatory factors,thereby alleviating pulmonary pathological damage.
