膝痹方对膝骨关节炎血瘀证大鼠MMP-13、ADAMTS-5表达的影响
Effects of Xibi Formula on MMP-13 and ADAMTS-5 Expression in Rats with Knee Osteoarthritis of Blood Stasis Syndrome

Abstract
Objective:To investigate the effects of Xibi Formula on the expression of matrix metalloproteinase-13(MMP-13)and a disintegrin and metalloproteinase with thrombospondin motifs-5(ADAMTS-5)in rats with knee osteoarthritis and blood stasis syndrome(KOA-BSS).Methods:Knee osteoarthritis was induced by intra-articular injection of papain and L-cysteine,while blood stasis syndrome was induced by epinephrine combined with ice-water stress.After successful modeling,rats were randomly divided into model group,positive control group,high-dose Xibi Formula group,and low-dose Xibi Formula group,with 8 rats in each group.Non-modeled rats receiving intra-articular and subcutaneous saline injections served as blank group.The high-and low-dose Xibi Formula groups received Xibi Formula decoction at doses of 50 g/kg and 25 g/kg(crude drug equivalent),respectively.The positive control group received diacerein(50 mg/kg),while the model and blank groups received saline,all administered by gavage for 6 weeks.General signs were observed and recorded before and after treatment.Knee joint function was evaluated using modified Lequesne MG index.After treatment,whole blood viscosity,plasma viscosity,and erythrocyte aggregation index were measured using an automatic hemorheology analyzer.Histopathological changes in articular cartilage were observed via hematoxylin-eosin staining and scored using Mankin's criteria.Plasma MMP-13 and ADAMTS-5 levels were detected by ELISA,while MMP-13 and ADAMTS-5 mRNA expression in cartilage was measured by RT-qPCR.Results:Compared with the model group,both Xibi Formula groups showed improved blood stasis symptoms,significantly reduced whole blood viscosity,plasma viscosity,and erythrocyte aggregation index(P<0.05,P<0.01),decreased Lequesne MG scores(P<0.01),improved cartilage surface smoothness,structural integrity,and chondrocyte arrangement,and lower Mankin's scores(P<0.05,P<0.01).The formula significantly downregulated plasma MMP-13 and ADAMTS-5 protein expression and cartilage MMP-13 and ADAMTS-5 mRNA levels(P<0.01).Conclusion:Xibi Formula effectively improves general signs and knee symptoms in KOA-BSS rats by downregulating cartilage degradation markers,demonstrating chondroprotective effects.关键词
