Study on the Anti-colon Cancer Effects of Flavonoid Extract from Sarcopyramis napalensis Wall in vitro

Abstract
Objective:To investigate the effects of flavonoid extract from Sarcopyramis napalensis Wall(FSN)on the proliferation and apoptosis of colon cancer HCT116 cells,and to explore its potential mechanisms.Methods:The CCK-8 assay was used to determine the effects of different concentrations of FSN on cell viability after 24 h and 48 h treatment of HCT116 cells.Based on the IC50 values,the experiments were divided into control group and low-,medium-,and high-concentration groups.Flow cytometry combined with Annexin V-FITC/PI double staining was employed to detect the effects of different concentrations of FSN on HCT116 cell apoptosis.Western blot was performed to examine the effects of different concentrations of FSN on the expression of apoptosis-related proteins Bax,Bcl-2,and Caspase-3 in HCT116 cells.Results:Different concentrations of FSN significantly inhibited the proliferation of HCT116 cells in a concentration-and time-dependent manner(P<0.05,P<0.01).After 24 h treatment with FSN,compared with the control group,the total apoptosis rates in the low-,medium-,and high-concentration groups were 14.34%,34.73%,and 40.93%,respectively,all significantly higher than that in the control group(P<0.01).Western blot results showed that compared with the control group,the expression of Bax and Caspase-3 was increased(P<0.05,P<0.01,P<0.001),while the expression of Bcl-2 was decreased(P<0.05,P<0.01)in the low-,medium-,and high-concentration groups.Conclusion:FSN can significantly inhibit the proliferation of colon cancer HCT116 cells and induce their apoptosis,possibly through up-regulating the expression of Bax and Caspase-3 while down-regulating the expression of Bcl-2.关键词
