Quality Evaluation of Entada phaseoloides Based on Multi-Index Quantification,Chemometrics,and Weighted TOPSIS Method

Abstract
Objective:To establish a multi-index quantitative detection method for Entada phaseoloides and explore the application of chemometrics and weighted TOPSIS method in its quality evaluation.Methods:A Nucleodur C18 Gravity column was used with a mobile phase of acetonitrile-0.2%phosphoric acid aqueous solution for gradient elution.The flow rate,detection wavelengths,and column temperature were set at 1.0 mL/min,280 and 210 nm,and 30℃,respectively.HPLC was employed to simultaneously determine the contents of 10 index components,alcohol-soluble extracts,total ash,and acid-insoluble ash in 18 batches of Entada phaseoloides.Using the multi-index content data as variables,chemometrics and weighted TOPSIS method were applied to construct a quality evaluation model.Results:Ten components were identified,namely entada saponin,entadamide A,entadamide A-β-D-glucopyranoside,catechin,eriodictyol,luteoloside,luteolin,stigmasterol,daucosterol,and β-sitosterol,with mass fractions ranging from 47.736 to 74.338,0.555 to 1.994,7.738 to 13.28,0.166 to 0.444,0.374 to 0.797,0.727 to 1.164,0.567 to 0.973,0.279 to 0.681,0.093 to 0.184,and 0.055 to 0.106 mg/g,respectively.The alcohol-soluble extract content ranged from 27.9%to 45.2%,total ash from 1.6%to 3.5%,and acid-insoluble ash from 0.1%to 0.5%,with certain fluctuations observed in each index.Chemometric analysis revealed that the 18 batches of Entada phaseoloides clustered into three categories,suggesting that entada saponin,entadamide A-β-D-glucopyranoside,eriodictyol,stigmasterol,and luteoloside are the main potential markers affecting product quality.Weighted TOPSIS analysis indicated that Entada phaseoloides from Guangdong and Guangxi exhibited better overall quality.Conclusion:The methods established using PCA,OPLS-DA,and weighted TOPSIS can be applied to evaluate the overall quality of Entada phaseoloides.关键词
