基于去势模型探讨骨痿方治疗骨质疏松症的实验研究
The Experimental Study on the Treatment of Osteoporosis with Guwei Prescription Based on Castration Model

Abstract
Objective:To investigate the bone protective effect and safety of the Chinese herbal compound"Guwei prescription"for tonifying the kidney and strengthening the spleen on castrated osteoporosis model mice.Methods:Bilateral ovariectomy was per-formed to establish an osteoporosis model in C57BL/6 female mice.Thirty 10-week-old C57BL/6 female mice were randomly di-vided into sham operation group(sham group),ovariectomy model group(OVX group)and a Guwei prescription group(GW group),with 10 mice in each group.Each group was gavaged on the first day after modeling.The GW group was given Guwei Fang decoction(2.16 g/kg·d),and the sham group and the OVX group were given an equal amount of 0.9%sodium chloride solution.The gavage was given once a day for 4 consecutive weeks.Micro-CT was used to analyze the bone volume fraction(BV/TV),trabecular number(Tb.N)and bone mineral density(BMD)of the proximal femur of mice;ELISA was used to detect the expression levels of bone metabolism indicators PINP and TRACP;HE staining of the heart,liver,spleen,lung,kidney and other major organs was used to evaluate the in vivosafety.Results:The BV/TV level in the OVX group was 54.5%lower than that in the Sham group(P<0.01),Tb.N was 65.2%lower(P<0.01),and BMD was 54.1%lower(P<0.01).The GW group significantly improved the above parameters(BV/TV increased by 36.2%,Tb.N increased by 45.5%,BMD increased by 29.2%,P<0.01).ELISA showed that the GW group re-versed the imbalance of bone metabolism induced by OVX(PINP increased by 24.0%,TRACP decreased by 21.9%,P<0.01).HE staining confirmed that there was no pathological damage to the heart,liver,kidney and other organs in the GW group.Conclusion:Guwei prescription can inhibit bone resorption,promote bone formation,significantly improve bone microstructure degeneration in ovariectomized mice,and reconstruct bone metabolic balance.It also has good in vivo safety at the experimental dose.关键词
