力克喘抗炎有效部位的筛选及其UPLC指纹图谱的构建
Screening of Anti-Inflammatory Active Fraction of Likechuan and Establishment of Its UPLC Fingerprint

Abstract
Objective:To screen the anti-inflammatory effective parts of Likechuan-inhaler and to establish the UPLC fingerprints of its effective parts.Methods:Aqueous extract and aromatic water were obtained by hydrodistillation and reflux extraction of the crude powder of Likechuan-preparation.The effect of aqueous extract of Likechuan-preparation on the survival rate of RAW264.7 cells was determined by MTT assay.An in vitro inflammation model was established by inducing RAW264.7 cells with lipopolysac-charide(LPS),then the changes in the level of Nitric Oxide(NO)content were detected by NO detection kit,and the protein expres-sion level of inducible Nitric Oxide Synthase(iNOS)was detected by immunoblotting.The quality of the anti-inflammatory effec-tive parts of 10 batches of Likechuan-preparation was analysed by UPLC method for evaluation,and the UPLC fingerprints of the extracts were established.Results:The RAW264.7 cells treated with different concentrations of Likechuan-preparation extracts showed no obvious trend of proliferation inhibition,and the RAW264.7 cells treated with the aqueous extract site showed a decrease in NO secretion at a concentration of 10 μg/mL,which showed a better anti-inflammatory effect.And it was further determined that the aqueous extract site by the low,medium and high administration doses of 100,200 and 400 μg/mL,could down-regulate the pro-tein expression of iNOS respectively.UPLC fingerprints were established for 10 batches of the aqueous extracts of Likechuan-preparation,and the similarities were 0.999,0.999,0.981,0.998,0.997,0.998,0.979,0.98,0.996,and 0.997,respectively.Conclu-sion:The components of Likechuan-preparation do not have any obvious cytotoxicity and the aqueous extract site is anti-inflammatory effective site.The similarity of the 10 batches of samples in this study is good,and the fingerprints established can pro-vide quality control and stable quality assurance of the herbs for subsequent experimental studies.关键词
