Based on PI3K/Akt Signaling Pathway,Research on Mechanism of"Three Methods and Three Points"Massage Manipula-tion in Improving Skeletal Muscle Atrophy

Abstract
Objective:To explore the effect of three methods and three points of massage on the gastrocnemius muscle of rats(SNI)injured by sciatic nerve clamp.Methods:18 SPF-grade SD male rats were randomly placed into sham operation(J)group,model(M)group and massage(T)group,6 rats/group.The SNI model was established by clamping the sciatic nerve of the right hind limb of SD rats in the M and T groups with special surgical forceps,while the sciatic nerve was only exposed in the J group without clam-ping.On the 7th day after operation,T group was interfered by the"intelligent massage manipulation simulator",and the massage ma-nipulation methods were simulated respectively on"Yinmen point""Chengshan point"and"Yanglingquan point",1 min/(point·method);J group and M group shall be held and bound.Once/d for 20 days.BBB(Basso,Beattie & Bresnahan locomotor rating scale)score and inclined plate test were performed before modeling and after 10 and 20 interventions.After 20 interventions,the rats were killed,and the gastrocnemius of the affected limb was taken,and the protein expressions of PI3 K,p-PI3 K,Akt and p-Akt in the gastrocnemius were detected by Western Blot.The mRNA expression of PI3K,Akt and FoxO3 was detected by RT-PCR.Results:①After 10 and 20 interventions,the BBB score and inclined plate test angle of M group and T group were lower than that of J group(P<0.05),and the BBB score and inclined plate test angle of T group were higher than that of M group(P<0.05).② After 20 in-terventions,the expressions of PI3K,Akt mRNA and P-PI3K and P-Akt mRNA protein in M group were lower than those in J group(P<0.05),while those in T group were higher than those in M group(P<0.05);The mRNA expression of FoxO3 in M group was higher than that in J group,and that in T group was lower than that in M group(P<0.05).Conclusion:"Three methods and three points"massage manipulation can improve the expression levels of p-PI3K,p-Akt and FoxO3 in the gastranemius muscle of SNI rats,and alleviate muscle atrophy by activating PI3K/Akt signaling pathway to up-regulate p-PI3K and p-Akt and down-regulate FoxO3,thereby reducing motor dysfunction caused by denervation of skeletal muscle atrophy.关键词
