健脑丸对C57BL/6 FMR1敲除小鼠行为学及海马神经元的影响
Effects of Jiannao Pill on Behaviorology and Hippocampal Neurons in C57BL/6 FMR1 KO Mice

Abstract
Objective:To investigate the effects of Jiannao Pill on the behaviorology and hippocampal neurons of C57BL/6 FMR1 knockout(KO)mice.Methods:Three female heterozygous C57BL/6 FMR1 KO mice were obtained via embryo recovery.After breeding with male wild-type homozygous C57BL/6 mice,C57BL/6 FMR1 KO mice and wild-type C57BL/6 mice were ob-tained.Ten wild-type C57BL/6 mice were selected as the wild-type group using a random number table.C57BL/6 FMR1 KO mice were divided into model group and Jiannao Pill group,with 10 mice in each group,also using a random number table.The Jiannao Pill group was given Jiannao Pill solution by gavage at a dose of 25 mL/(kg·d)based on body weight,while the wild-type and model groups were gavaged with an equal volume of pure water.All three groups were intervened for 28 days.Using CatWalk gait analyzer to evaluate and record behavioral parameters from three dimensions:foot trajectory,overall movement level,and limb movement lev-el.The overall movement level includes average speed,crawling distance,number of steps and step frequency;the level of limb move-ment includes swing time,step distance,support time,and maximum contact area;three groups of mice were scanned using magnetic resonance imaging(MRI)and their brain volumes were compared.After cardiac perfusion,the brains were weighed and the brain mass was compared;the brain tissue of mice was stained using the Nissl staining method,and the number of neurons in the hippocam-pal CA1 to CA4 regions was observed and counted under a light microscope.Results:Compared with the model group,the mice in the wild-type and Jiannao Pill groups had uniform step length and more regular footsteps.And their average speed,crawling distance,number of steps,step frequency,swing time,and step distance all increased(P<0.05),while the support time and maximum contact area both decreased(P<0.05);there was no statistically significant difference in brain volume and brain mass among the three groups of mice(P>0.05);the number of neurons in the CA1 and CA3 regions of the mice in wild-type and Jiannao Pill groups were higher than that in the model group(P<0.05),while there was no statistically significant difference in the number of neurons in the CA2 and CA4 regions(P>0.05).Conclusion:Jiannao Pill can significantly improve the motor ability of C57BL/6 FMR1 KO mice,promote an increase in the number of hippocampal neurons and structural repair in the brain.关键词
