基于HIF-1α/VEGF信号通路探讨逐瘀汤对子宫内膜异位症模型大鼠的影响
Effects of Zhuyu Decoction on Endometriosis Rats Based on HIF-1α/VEGF Signaling Pathway

Abstract
Objective:To investigate the effects of Zhuyu Decoction on HIF-1α/VEGF signaling pathway in rats with endometrio-sis(EM).Methods:18 endometriosis model rats were established using autologous uterine transplantation.After 3 weeks,the success-fully modeled rats were randomly divided into model group,Gestrinone group,and Zhuyu Decoction group according to a random number table,with 6 rats in each group.Additionally,6 normal SD rats were selected as the sham surgery group.The sham surgery and model groups were given 10 mL/(kg·d)of physiological saline by gavage of 3 mL,the Gestrinone group was given 0.5 mg/(kg·d)of gestrinone by gavage of 3 mL,and the Zhuyu Decoction group was given 10 mL/(kg·d)of Zhuyu Decoction by gavage of 3 mL.After 28 days of continuous administration,the morphology and volume changes of ectopic lesions in 4 groups of rats were observed.HE staining was used to observe the pathological changes of ectopic lesions in 4 groups of rats,Western blot was used to detect the protein expressions of HIF-1α and VEGF and immunohistochemistry was applied to determine the expressions of HIF-1α and VEGF positive cells in ectopic lesion tissues of rats in 4 groups.Results:There was no ectopic lesion formation in the sham surgery group.Compared with the sham surgery group,the model group showed typical ectopic lesions with vesicular structures,and typical endome-trial tissue morphology was observed within the ectopic lesions.Compared with the model group,the ectopic lesions in the Zhuyu De-coction and Gestrinone groups also appeared pale pink with typical vesicular structures,and their volumes reduced compared to the model group(P<0.05).There was no significant change in the morphology of ectopic lesions between the Gestrinone group and Zhuyu Decoction group,and the difference in volume ratio between the two groups was not statistically significant(P>0.05).The HE staining results showed that compared with the sham surgery group,the model group rats had neutrophil aggregation,increased glan-dular and stromal cells,inflammatory cell infiltration,and abundant blood vessels in the ectopic lesion vesicles.The Zhuyu Decoction and Gestrinone groups showed significant reduction in columnar epithelial cell shedding,interstitial looseness,neovascularization,and inflammatory cells.Compared with the sham surgery group,the expression levels and ratio of positive cells of HIF-1α and VEGF proteins in ectopic lesion tissues of rats in the model group increased(P<0.05).Compared with the model group,the expression levels and ratio of positive cells of HIF-1α and VEGF proteins in ectopic lesions reduced in the Gestrinone and Zhuyu Decoction groups(P<0.05).There was no statistically significant difference in the above indicators between the Gestrinone group and the Zhuyu Decoction group(P>0.05).Conclusion:Zhuyu Decoction can inhibit the protein expression of HIF-1α and VEGF in the VEGF-related signaling pathway of ectopic endometrium in rats with endometriosis.关键词
