摘要

Abstract

Carnitine palmitoyl transferase 1A(CPT1A)is the rate-limiting enzyme of mitochondrial fatty acid β oxidation and plays a key role in energy metabolism.The full-length CDS sequence of chicken CPT1A gene was obtained by RT-PCR,and the cloning vector of the gene was successfully constructed by directional cloning.The differential expression of the gene in 16 tissues and cells such as chicken abdominal fat,muscle stomach fat,Immortalized chicken preadipocyte cell 1 and Intramuscular preadipocytes was detected by RT-QCPR.The protein structure,subcellular localization and phylogenetic relationship were predicted by bioinformatics tools.The results showed that the total length of chicken CPT1A gene ORF was 2 313 bp,encoding 770 amino acids.CPT1A protein has no signal peptide and has two transmembrane domains,and subcellular localization is in cytoplasm and mitochondrial membrane,so it belongs to hydrophilic stable protein.The secondary structure mainly exists in the form of α helix.Phylogenetic tree shows the closest relationship with birds.The study provides a molecular basis for the functional study of CPT1A in chicken lipid metabolism.
