Abstract

To investigate whether baicalein could inhibit BVDV replication by inhibiting autophagy induced by BVDV infection in vitro and in vivo,mRNA and protein expression of autophagy-related genes were detected by RT-qPCR and Western blot,and changes of autophagosomes in the spleen of mice were observed by transmission electron microscopy.The results showed that when BVDV-infected cells were treated with the autophagy inducer rapamycin(RAPA)in this study,baicalein effectively inhibited the rapamycin-induced up-regulation of Atg12,Beclin-1 and LC3B mRNA expression and the down-regulation of p62 mRNA expression,and decreased the expression levels of p-PI3K,p-Akt,and p-mTOR;baicalein had a positive effect on the PI3K/Akt/mTOR signaling pathway,suggesting that it may play a role in the abnormal cell signaling pathway induced by BVDV infection.Baicalin inhibited the abnormal mTOR signaling pathway as well as BVDV-induced cellular autophagy,inheriting BVDV replication in vitro and in vivo,and the results of the study provide a new idea for the treatment of BVDV infection.
