基于BVDV诱导的自噬和mTOR信号通路的黄芩苷体内外抗BVDV作用
Anti-BVDV Effects of Baicalin Based on BVDV-Induced Autophagy and mTOR Signaling Pathway in vitro and in vivo

Abstract
To investigate whether baicalein could inhibit BVDV replication by inhibiting autophagy induced by BVDV infection in vitro and in vivo,mRNA and protein expression of autophagy-related genes were detected by RT-qPCR and Western blot,and changes of autophagosomes in the spleen of mice were observed by transmission electron microscopy.The results showed that when BVDV-infected cells were treated with the autophagy inducer rapamycin(RAPA)in this study,baicalein effectively inhibited the rapamycin-induced up-regulation of Atg12,Beclin-1 and LC3B mRNA expression and the down-regulation of p62 mRNA expression,and decreased the expression levels of p-PI3K,p-Akt,and p-mTOR;baicalein had a positive effect on the PI3K/Akt/mTOR signaling pathway,suggesting that it may play a role in the abnormal cell signaling pathway induced by BVDV infection.Baicalin inhibited the abnormal mTOR signaling pathway as well as BVDV-induced cellular autophagy,inheriting BVDV replication in vitro and in vivo,and the results of the study provide a new idea for the treatment of BVDV infection.关键词
