山东省部分地区水貂阿留申病PCR检测及病原的分离鉴定
PCR Detection,Pathogen Isolation and Identification of Mink Aleutian Disease in Some Areas of Shandong Province

Abstract
In order to investigate the prevalence and biological characteristics of isolated strains of mink Aleutian disease virus(ADV)in Shandong Province,specimens were collected from five major mink breeding areas in Shandong Province,for PCR detection,isolation and cultivation of ADV,analysis of homology and genetic variation of isolated strains.The results showed that 1 917 bp specific band of ADV VP2 gene was obtained by PCR using ADV-specific primers.The positive rate of ADV in the mink farms of the five regions was 38.9%-67.1%.At the same time,a new strain of ADV,named ADV-SD1908,was isolated and identified,which exhibited significant cytopathic effects(CPE)on Crandell Reese Feline Kidney(CRFK)cells.TCID50 of the isolated strain was 10-3.64/mL.The homology between this strain and other strains is 94.4%-96.1%,and it has the closest genetic relationship with domestic HLJ strain and foreign Pullman strain.The research results will provide references for the epidemiology,genetic evolution,prevention and control of mink ADV in Shandong Province.关键词
