Abstract

This study aimed to apply juvenile in vitro embryo transfer(JIVET)technology to Ordos fine-wool sheep to accelerate the introduction of prolificacy genes and breeding progress.Ten 5~6-week-old female lambs were selected as donors.After hormonal induction and ovum pickup,the collected oocytes underwent in vitro maturation and were fertilized in vitro with semen from prolific fine-wool rams.A portion of embryos developed to blastocyst and morula stages were transferred to recipient ewes.Genotyping was performed on lambs after birth.Results showed:649 oocytes were collected from 10 donor lambs,of which 534 were subjected to fertilized in vitro after in vitro maturation,yielding 331 embryos.Selected blastocysts/morulae were transferred to 74 recipients,resulting in 16 live lambs.Donor lambs weighing>3.6 kg yielded significantly fewer oocytes(mean:14.0)compared to other groups(83.3 and 85.5,respectively).Pregnancy rates were higher for rapidly developing embryos(Day 6 blastocysts)than for slower-developing ones(Day 7 blastocysts/morulae).All 16 offspring lambs produced carried the FecB gene,including seven homozygotes(43.8%,7/16)and nine heterozygotes(56.2%,9/16).This study demonstrates that combining JIVET with molecular marker technology accelerates FecB gene introduction in Ordos fine-wool sheep,providing a feasible approach to shorten breeding cycles and develop new prolific strains.
