Abstract

This study aimed to evaluate the safety of the recombinant Lactobacillus strain LP3-2 expressing the F protein of Newcastle disease virus(NDV).LP3-2 was cultured on MRS agar plates,followed by Gram staining and microscopic examination to observe colony and cellular morphology.Hemolytic activity was assessed via blood agar plate culture,and antibiotic susceptibility to seven common antibiotics was determined using the minimum inhibitory concentration(MIC)method.Mice were infected by oral gavage(5.0×107~5.0×109 CFU/0.2 mL per mouse daily for 28 days),and the animal pathogenicity of strain LP3-2 was analyzed.Results showed no hemolysis on blood agar plates,and LP3-2 exhibited sensitivity to all tested antibiotics.None of the mice that received oral administration of the bacterial suspension solution showed obvious clinical symptoms.There were no visible lesions in the heart,liver,spleen,lungs or kidneys of the mice.The serum creatinine(CREA)content of mice in each experimental group was significantly lower than that in the control group(P<0.05).The study demonstrates that this recombinant Lactobacillus expressing NDV F protein has favorable safety profiles,providing a foundation for its further application in novel vaccine development.However,long-term safety and potential risks require further in-depth research.
