光甘草定纳米粒子的制备及其对帕金森病小鼠运动功能影响
Preparation of glabridin nanoparticle and its effect on motor function in mice with Parkinson's disease

Abstract
Objective To prepare glabridin nanoparticle,and to investigate its effect on motor function in mice with Parkin-son's disease(PD)induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP).Methods Glabridin and polydopa-mine(PDA)were used to synthesize glabridin nanoparticles(Glab-PDA NPs)through self-assembly,and then Glab-PDA NPs were characterized.A total of 24 C57BL/6 mice,aged 8 weeks,were randomly divided into MPTP group,Glab-PDA NPs treat-ment group,and control group,with 8 mice in each group.The mice in the MPTP group and the Glab-PDA NPs treatment group were given intraperitoneal injection of MPTP to establish a model of subacute PD.The mice in the Glab-PDA NPs treatment group were given injection of Glab-PDA NPs through the caudal vein since day 3 before modeling till the end of modeling,while those in the control group were injected with an equal volume of normal saline.The rotarod test and the gait test were used to assess the motor ability of mice.Results The homogeneous round Glab-PDA NPs were prepared successfully,with a drug encapsulation efficiency of 40%and a drug loading content of 8.7%.The rotarod test showed that the MPTP group had a significantly worse mo-tor coordination ability than the control group(F=5.97,q=2.778,P<0.05),while the Glab-PDA NPs treatment group showed significant recovery of motor function(q=3.211,P<0.01).The gait test showed that the MPTP group had significant reductions in the motor ability of the limbs(F=3.03-52.82,q=2.374-9.521,P<0.05),while the Glab-PDA NPs treatment group had sig-nificant improvements in the indicators of the limbs(q=2.826-8.198,P<0.05).Conclusion Glab-PDA NPs can improve mo-tor function and restore motor ability in PD mice.关键词
