超声对PC12细胞的调控及对6-OHDA诱导的帕金森病小鼠模型运动行为的影响
Regulatory effects of ultrasound on PC12 cells and motor behaviors of mouse model of 6-OHDA-induced Parkinson's disease

Abstract
Objective To investigate the effects of ultrasound(US)on pheochromocytoma(PC12)cells and the motor be-haviors of a mouse model of 6-hydroxydopamine(6-OHDA)induced Parkinson's disease.Methods PC12 cells were divided into control group and US group(stimulated with US),and the Ca2+fluorescence intensity of the cells in the two groups was measured.C57BL/6J mice were divided into control group,model group,and US group to receive injection of normal saline,6-OHDA solu-tion,and 6-OHDA solution into the striatum,respectively;after 2 weeks,gait analysis was performed for the three groups;and then,the US group was given US stimulation for 3 d,followed by gait analysis for the locomotion ability of the mice.Results PC12 cells in the US group showed a significant increase in the fluorescence intensity of Ca2+than those in the control group(t=3.071,P<0.05).Compared with the model group,the mice in the US group showed a significantly larger maximum contact area of the paws(F=2.99,q=2.410,P<0.05)and a significantly higher movement frequency(F=40.35,q=3.879,P<0.01).In the US group,the step length of the mice was increased significantly after US stimulation compared with before US stimulation(t=2.636-9.760,P<0.05).The content of tyrosine hydroxylase(TH)in the substantia nigra(SN)of the mice in the US group was significantly increased than that in the model group(F=78.03,q=3.589,P<0.05)but significantly lower than that in the control group(q=9.821,P<0.001).Conclusion US can promote Ca2+inflow in PC12 cells,alleviate the movement disorder of PD mice,and save TH-positive neurons in the SN region.关键词
