ACSL4对MPP+处理后N2a细胞线粒体数量的影响
Effect of acyl-CoA synthetase long-chain family member 4 on mitochondrial number in N2a cells after 1-methyl-4-phenylpyridinium treatment

Abstract
Objective To investigate the effect of acyl-CoA synthetase long-chain family member 4(ACSL4)on mitochon-drial number in N2a cells after 1-methyl-4-phenylpyridinium(MPP+)treatment.Methods Western blotting was used to mea-sure the protein expression level of ACSL4 in N2a cells after MPP+treatment.N2a cells were transfected with Flag-Vector or Flag-ACSL4 plasmids,and Western blotting was used to observe the effect of ACSL4 overexpression on the expression levels of voltage-dependent anion channel 1(VDAC1)and mitochondrial import receptor subunit TOM20 homolog(TOMM20)in N2a cells after MPP+treatment.Results Compared with the control group,the MPP+treatment group had a significant increase in the protein expression level of ACSL4 in N2a cells(t=3.965,P<0.05).Compared with the control group,the MPP+treatment group had significant increases in the expression levels of VDAC1 and TOMM20(F=28.59,15.15,P<0.01),while compared with the MPP+treatment group,ACSL4 overexpression had no further impact on the expression levels of VDAC1 and TOMM20 in N2a cells(P>0.05).Conclusion ACSL4 overexpression has no impact on mitochondrial number in N2a cells after MPP+treat-ment,suggesting that the role of ACSL4 in the pathogenesis of Parkinson's disease does not involve its impact on mitochondrial number.关键词
