MPTP和α-Syn预制原纤维诱导的慢性帕金森病模型小鼠黑质多巴胺能神经元的自发放电活动
Spontaneous discharges of nigral dopaminergic neurons in mouse models of chronic Parkinson's disease induced by MPTP and α-Syn PFFs

Abstract
Objective To investigate the spontaneous activity of substantia nigra dopaminergic neurons in two mouse models of chronic Parkinson's disease induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)and alpha-synuclein preformed fibrils(α-Syn PFFs),respectively.Methods Two mouse models of chronic Parkinson's disease were prepared by long-term in-jection of MPTP into the peritoneal cavity and stereotaxic injection of α-Syn PFFs into the right striatum of the brain,respectively.We recorded the spontaneous firing activity of dopaminergic neurons in the substantia nigra pars compacta of the two models using electrophysiology techniques.Results The MPTP group and α-Syn PFFs group showed significantly smaller mean numbers of firing dopaminergic neurons per recording session in the substantia nigra pars compacta(t=3.015,4.779,both P<0.05),signifi-cantly higher firing rates of nigral dopaminergic neurons(t=-3.070,-4.016,both P<0.05),and significantly higher propor-tions of nigral dopaminergic neurons exhibiting burst firing(t=-3.611,-2.928,both P<0.05),compared to the control group.Conclusion Mouse models of chronic Parkinson's disease induced by MPTP and α-Syn PFFs differ significantly from control mice,mainly manifesting with decreased numbers of neurons exhibiting spontaneous firing and increased firing rates and proportions of neurons exhibiting burst firing.关键词
