Effect of 1-methyl-4-phenylpyridinium-induced inflammation on the expression of Dectin-1 and inducible nitric oxide synthase in BV2 microglial cells

Abstract
Objective To investigate the effect of 1-methy-4-phenylpyridinium(MPP+)-induced inflammation on the ex-pression levels of Dectin-1 and inducible nitric oxide synthase(iNOS)in BV2 microglial cells.Methods BV2 microglial cells were treated with different concentrations of MPP+(0.1,0.5,and 1.0 mmol/L),and MTT assay was used to measure cell viabili-ty.Western blotting was used to observe the effect of different durations of MPP+treatment(2,4,6,12,and 24 hours)on the protein expression levels of the pattern recognition receptor Dectin-1 and the inflammatory factor iNOS in BV2 microglial cells.Results MPP+at the concentration of 0.5 mmol/L and 1.0 mmol/L significantly reduced the viability of BV2 microglial cells(F=9.058;q=3.611,4.361;P<0.05).There was a significant increase in the protein expression level of Dectin-1 at 4,6,and 12 hours after MPP+induction(F=5.660,q=4.784-6.610,P<0.05),which returned to normal at 24 hours.There was a signi-ficant increase in the protein expression level of iNOS at 12 and 24 hours after MPP+treatment(F=4.884;q=3.662,3.931;P<0.05).Conclusion MPP+-induced inflammatory response in BV2 microglial cells can significantly increase the protein expres-sion of the pattern recognition receptor Dectin-1 and iNOS.关键词
