Abstract

To explore the genetic diversity of Panus giganteus germplasm resources and accelerate the breeding process of high-quality mushroom varieties,10 strains of P.giganteus were used as experimental materials to measure their agronomic traits.Antagonistic tests and molecular marker analyses using ISSR,RAPD,and SRAP were conducted to assess genetic di-versity.DNA fingerprinting was performed based on the amplification results of the three molecular markers.Cluster analysis of agronomic traits revealed that the 10 strains were divided into four groups at a Euclidean distance of 2.5.The 8 ISSR prim-ers,10 RAPD primers,and 12 pairs of SRAP primers amplified 111,121,and 166 clear bands,respectively,with polymorphism ratios of ISSR(81.98%)>SRAP(77.71%)>RAPD(66.12%).Comprehensive clustering based on three molecular markers re-vealed that the 10 strains were divided into four groups at a genetic similarity coefficient of 0.76,with strains of similar ge-netic backgrounds clustering together.The tested strains exhibited rich genetic diversity,and the clustering results based on ISSR,RAPD,and SRAP markers were consistent with those of agronomic traits and antagonistic tests.ISSR molecular mark-ers are more consistent with the genetic background and can be used for DNA fingerprinting.These findings provide evidence for the identification and genetic relationship analysis of P.giganteus.
