摘要

Abstract

Using the intact,crack-free capsules of wild Dendrobium aphyllum from Lincang,Yunnan Province,as the mate-rial,this study investigated aseptic germination,protocorm induction,tufted bud induction and proliferation,rooting and ac-climatization,and transplanting.The growth of Dendrobium aphyllum was compared on aseptic germination medium,aseptic seedling proliferation medium,and tufted bud rooting medium,supplemented with different concentrations of organic addi-tives,inorganic salts,and plant growth regulators.The optimal medium formula for each culture stage was selected to establish an in vitro rapid propagation system for D.aphyllum.The results revealed that the optimal medium for aseptic seed germina-tion was MS+potato juice(60 g·L-1)+sucrose(30 g·L-1)+agar(6.0 g·L-1).The optimal medium for tufted bud proliferation was MS+potato juice(60 g·L-1)+6-BA(0.5 mg·L-1)+NAA(0.2 mg·L-1)+AC(0.2 g·L-1),with a proliferation coefficient of 4.6.The optimal medium for robust seedling and rooting was 1/2 MS+banana juice(60 g·L-1)+IBA(0.5 mg·L-1)+6-BA(0.2 mg·L-1)+NAA(1.0 mg·L-1)+AC(0.2 g·L-1).Seedlings exhibited robust growth and well-developed root systems,with an average plant height of 3.28 cm,an average number of roots per plant of 15.6,and an average root length of 2.15 cm.The survival rate reached 100% after 30 days of acclimatization and transplanting.
