川西獐牙菜SmCHI1基因克隆及表达分析
Cloning and expression analysis of SmCHI1 gene in Swertia mussotii Franch

Abstract
Objective The gene structure information of chalcone isomerase(CHI)gene in Swertia mussotii Franch.and its expression regulation mode in flavonoid biosynthesis pathway were described.Methods The specific primers were designed to clone the SmCHI1 gene from S.mussotii.The bioinformatics analysis was carried out to construct the prokaryotic expression vector,and the expression characteristics were analyzed by RT-qPCR.Results The open reading frame of SmCHI1 is 660 bp in length.SmCHI1 protein has chalcone cyclization activity.The SmCHI1 recombinant protein was induced by IPTG,and it was found that the concentration of IPTG had no significant effect on the expression of SmCHI1 protein.Tissue-specific expression analysis showed that SmCHI1 was highly expressed in roots and leaves.Conclusion The results of this study can lay a foundation for further study on the function of SmCHI1 in the synthesis pathway of flavonoids in S.mussotii.关键词
