白术多糖的初步结构表征及生物活性研究
Study on the preliminary structural characterization and bioactivities of polysaccharides from Atractylodes macrocephala Koidz

Abstract
Objective The aim of this study was to investigate the optimised extraction method,structural characterization,antioxidant activity and immunological activity of polysaccharides from Atractylodes macrocephala Koidz..Methods The polysaccharide content was determined by phenol sulfuric acid method,and the optimal extraction method of Atractylodes macrocephala Koidz.was investigated using the polysaccharide content as an index;The functional groups,molecular weights and monosaccharide compositions were determined by infrared spectrometry,HPGPC,and PMP-HPLC,respectively;The antioxidant activities of were probed by DPPH·and·OH radicals;RAW264.7 cells were used to investigate the immunological activity by MTT method and flow cytometry.Results The optimal extraction method for Atractylodes macrocephala polysaccharide was hot water extraction;Infrared spectroscopy results showed that Atractylodes macrocephala polysaccharide contained O-H,C-H and C-O bond,etc.,and had α and β configuration;HPGPC results showed that Atractylodes macrocephala polysaccharide was a homogeneous polysaccharide with a molecular weight of 3 011.5 Da;PMP-HPLC results showed that Atractylodes macrocephala polysaccharide was composed of Man,Rha,GalA,Glc,Xyl,and Ara with a molar ratio of 1.00∶1.38∶4.88∶51.88∶6.88∶8.00;Atractylodis macrocephalae polysaccharide showed a certain antioxidant activity with better DPPH·and·OH-scavenging effects;Furthermore,they significantly promoted the proliferation and phagocytosis activity of RAW264.7 cells,and possessed significant immunological activity.Conclusion A homogeneous polysaccharide of Atractylodes macrocephala could be obtained by hot water extraction and showed significant antioxidant and immunological activities.关键词
