摘要

Abstract

In order to study the specific role of DlWRKY3 gene in response to abiotic stresses,the nucle-otide sequence of DlWRKY3 gene was cloned using homologous cloning method with the nucleotide sequence of DkWRKY3 gene of Diospyros kaki as reference.Bioinformatics and expression analyses of DlWRKY3 gene were performed.The results showed that the length of open reading frame(ORF)of DlWRKY3 gene was 951 bp,encoding 316 amino acids.The molecular formula of DlWRKY3 protein was C1490H2401N447O490S15,and the the-oretical isoelectric point was 8.52.It was an unstable alkaline hydrophilic protein with not transmembrane do-main or signal peptide region.The predicted secondary structure of this protein was dominated by α-helix and random curling.It was a type Ⅱ WRKY transcription factor with the typical WRKYGQK domain and a C2H2 zinc finger structure.Homologous comparison showed that this gene was closely related to Arabidopsis At-WRKY40 and cotton GhWRKY3.The results of fluorescence quantitative PCR indicated that the expression of DlWRKY3 was induced by low temperature stress,and inhibited by drought and salt stresses.In this study,Dl-WRKY3 gene was successfully cloned,which laid a foundation for further study on the role of DlWRKY3 gene in response to abiotic stresses,and provided the important gene resource and theoretical basis for stress-resist-ant molecular breeding of Diospyros plants.
