p90RSK在脓毒症内皮功能障碍与凝血紊乱中的作用
The role of p90RSK in endothelial dysfunction and coagulation regulation in sepsis

Abstract
Objective This paper reports that the inhibition of phospho-Ser380 of p90RSK can be an effective means of treating sepsis through endothelial dysfunction and coagulation regulation.Methods In vivo,mice were performed to cecal ligation and puncture(CLP)surgery to simulate a sepsis model and then assessed for endothelial function,coagulation cascades,and survival.In vitro,HUVEC cells were exposed to lipopolysaccharide(LPS)to establish cellular models of sepsis.The expression of endothelial function,coagulant factors,and signaling pathways were detected.Further-more,the ERK/p90RSK pathway was validated using FMK.Results FMK attenuates the activation of the septic coagulation cascade,suppresses the secretion of coagulation-related mediators,alleviates tissue injury,and subsequently reduces mortality in septic mice.In addition,the secretion of the phosphorylation of p90RSK in HUVEC cells exposed to LPS stimulation was attenuated by administe-ring a p90RSK inhibitor.Conclusions This study suggested that FMK may inhibit the LPS-induced triggering of the ERK/p90RSK pathway and down-regulate PAI-1 and TF expression,subsequently a-meliorating the coagulation imbalance and endothelial dysfunction in the sepsis state.关键词
