基于KASP技术的SNP标记用于3个松花菜新品种种子纯度检测
Application of SNP Marker Based on KASP Technology in Seed Purity of Three New Varieties of Loose-curd Cauliflower

Abstract
To address the intense competition in the seed market and ensure the robust development of the cauliflower seed industry,genotyping technology was employed for precise and efficient variety identification and purity assessment of novel loose cauliflower cultivars.Single nucleotide polymorphisms(SNPs)were detected using the competitive allele-specific Kompetitive Allele Specific PCR(KASP PCR)technique on loose cauliflower as the experimental material.High-throughput sequencing facilitated the identifica-tion of SNP loci suitable for broccoli variety discrimination through bioinformatics analysis.Subsequently,the purity of the new Songhua varieties Jinsong 75,Jinsong 90,and Jinsong 105 was assessed using the identified SNP loci and KASP genotyping technolo-gy.The results demonstrated a high level of agreement between the genotyping results and purity assessment(r≈ 0.904).In conclu-sion,the SNP marker-based approach established in this study offers a reliable method for variety identification and purity assess-ment,thereby mitigating economic losses from misidentified hybrids,minimizing potential disputes,enhancing identification efficiency and accuracy,providing substantial evidence for variety protection,and presenting significant practical value in the realm of Songhua hybrid identification and variety safeguarding.关键词
