摘要

Abstract

In order to establish a rapid propagation system for Cornus florida,tissue culture methods were used.The study involved screening sterilization methods for stem segment explants of Cornus florida and investigating the effects of different concentrations of hormones NAA and 6-BA on callus induction,bud differentiation,young leaf proliferation,and rooting.The results indicated that ster-ilization with 0.1%HgCl2 for 12 minutes was optimal.Callus induction was most effective with MS+1.0 mg·L-1 6-BA+0.2 mg·L-1 NAA,reaching a 90%induction rate after 40 days.Bud differentiation from callus was best promoted by MS+2.0 mg·L-1 6-BA+0.2 mg·L-1 NAA.Shoot proliferation from sterile young leaves was highest with MS+1.0 mg·L-1 NAA+0.5 mg·L-1 6-BA.Root induction was best in MS+0.5 mg·L-1 IBA,however,the induced roots were prone to browning,necessitating timely subculture onto a rejuvena-tion medium In condusion,this study provides a technical foundation for the large-scale propagation of this species,which is of great significance for its resource development and industrial application.
