Establishment of Tissue Culture and Regeneration System of Clematis huchouensis Tamura,an Endemic Plant in China

Abstract
In order to realize industrial production and accelerate the popularization and application process,it is the best way to establish the plant tissue culture system of Clematis huchouensis.In this study,two-year-old stem segments of clematis from Xing Wu with buds were used as explants for tissue culture and rapid propagation.The results showed that the mixed treatment of 70%alcohol sterilization for 10 s and 10%sodium hypochlorite sterilization for 10 min was the best,with a germination rate of 70.67%and a pollution rate of 17.33%.1/2MS medium is more suitable for start-up culture,and the sprouts grow well,not only robust,but also without yellowing and browning.In the multiplication culture,MS+2 mg·L-16-BA+0.3 mg·L-1 IBA+2 g·L-1 hydrolyzed casein was the best,and the multiplication coefficient was 4.73.The best rooting culture medium was 1/2 ms+0.5 mg·L-1 IBA+0.01 mg·L-1 NAA,and the rooting rate was 79.67%.This study established a complete tissue culture technology system for Clematis huchouensis,breaking its geographical limitations.It provides a theoretical basis for its promotion and commercial development,and also offers cer-tain support for its interdisciplinary cooperation in the later stage.关键词
