Abstract

Objective It is to investigate the effect of Sanbaohutong decoction on the immune imbalance of Th17/Treg cells in rats with bronchial asthma with syndrome of lung-spleen qi deficiency.Methods Sixty male SD rats aged 2-3 weeks were randomly divided into control group,model group,montelukast sodium group,and low-,medium-,and high-dose groups of Sanbaohutong decoction,with 10 rats in each group.The rats of all groups except for the control group were sensi-tized via intraperitoneal injection of ovalbumin(OVA)and aluminum hydroxide,followed by nebulized inhalation of OVA,and induced by factors of lung-spleen qi deficiency at the same time to establish models of bronchial asthma with syndrome of lung-spleen qi deficiency.After successful modeling,the low-,medium-,and high-dose groups of Sanbaohutong decoc-tion were given Sanbaohutong decoction 3.57 g/kg,7.14 g/kg,and 14.28 g/kg by gavage,respectively;the montelukast sodium group was given montelukast sodium 0.525 mg/kg by gavage,while the control group and model group were given an equal volume of physiological saline by gavage,all once daily,continuously treated for 14 days.The general condition of the rats was observed and their body weight was recorded,the pathological morphology of lung tissue was observed by HE staining,the serum levels of interleukin-17(IL-17),transforming growth factor-β(TGF-β),interleukin-6(IL-6),signal transducer and activator of transcription 3(STAT3),interleukin-10(IL-10)were measured by ELISA,and the mRNA ex-pressions of retinoic acid-related orphan receptor γt(RORγt)and forkhead box protein P3(FoxP3)in lung tissue were de-tected by RT-PCR.Results After 14 days of gastric lavage,the body weight of rats in the model group was significantly lower than that of the control group(P<0.05);the body weight of rats in all groups of Sanbaohutong decoction was signifi-cantly higher than that of the model group(P<0.05),and the high-dose group of Sanbaohutong decoction was significantly higher than the low-and medium-dose groups of Sanbaohutong decoction and montelukast sodium group(all P<0.05).Compared with the control group,the bronchial lumens of rats in the model group were narrowed with a extensive inflamma-tory cell infiltration around the airways;the serum levels of IL-17,TGF-β,IL-6,and STAT3,as well as the relative ex-pressions of RORγt mRNA in lung tissue were significantly increased(all P<0.05),while the serum level of IL-10 and the relative expressions of FoxP3 mRNA in lung tissue were significantly reduced(all P<0.05).Compared with the model group,the lung histopathological injury in all groups of Sanbaohutong decoction and montelukast sodium group were signifi-cantly alleviated,the serum levels of IL-17,TGF-β,IL-6,and STAT3,as well as the relative expressions of RORγt mR-NA in lung tissue were significantly reduced(all P<0.05),while the serum level of IL-10 and the relative expressions of FoxP3 mRNA in lung tissue were significantly increased(all P<0.05).Conclusion Sanbaohutong decoction can improve the symptoms of bronchial asthma and alleviate the pathological damage of lung tissue in rats with bronchial asthma with syndrome of lung-spleen qi deficiency,the mechanism maybe related to regulating Th17/Treg immune balance.
