线粒体自噬在糖尿病心肌病中的作用及其研究进展
Role and research progress of mitophagy in diabetic cardiomyopathy
摘要
Abstract
Diabetic cardiomyopathy(DCM)is a non-ischemic,non-hypertensive myocardial disorder induced by metabolic disturbances in patients with diabetes mellitus(DM),and it significantly affects prognosis and mortality.As the central organelle for cellular energy metabolism,mitochondrial dysfunction represents a key pathological mechanism in DCM.Mitophagy,as a critical component of mitochondrial quality control,plays a dual regulatory role in DCM progression by selectively removing damaged or dys-functional mitochondria.This review systematically summarizes the molecular mechanisms of mitophagy,including ubiquitin-dependent pathways(PINK1/Parkin pathway),receptor-mediated pathways(FUNDC1,BNIP3/NIX,PHB2,etc.),and pathways mediated by TFEB/calcineurin.It also explores their roles in DCM(such as time-dependent changes in autophagy in HFD models,potential detri-mental effects of enhanced autophagy in T1DM)and synthesizes research progress on how antidiabetic drugs(empagliflozin,liraglu-tide,metformin,etc.)improve DCM through mitophagy regulation.Elucidating the regulatory mechanisms and targeted intervention strategies of mitophagy may provide novel theoretical foundations and therapeutic directions for DCM prevention and treatment.关键词
