血液代谢物与脑梗死的因果关系:一项两样本双向孟德尔随机化研究
Causal relationship between blood metabolites and cerebral infarction:a two-sample two-way Mendelian randomization study

Abstract
Objective To explore the causal relationship between blood metabolites and cerebral infarction(CI)by two-sample two-way Mendelian randomization(MR)analysis.Methods The published genome-wide association studies(GWAS)data of cerebral infarction were obtained from public database.With a GWAS containing 1400 circulating metabo-lites as an exposure indicator,SNPs which strongly related to exposure factors were selected as instrumental variables(IV).The causal relationship between 1400 blood metabolites and cerebral infarction was analyzed by R4.3.2 software.Five meth-ods—MR-Egger regression,inverse-variance weighted(IVW),weighted median,simple mode,and weighted mode—were employed to assess heterogeneity,pleiotropy,and sensitivity,thereby verifying the robustness of the results.Finally,a reverse MR analysis was performed on the obtained results.Results IVW method was used to assess causal relationship between blood metabolites and CI,and the results showed that urea(OR=0.997,95%CI:0.996-0.999)was a protec-tive factor,and the risk of CI decreased with the increase of metabolite urea.The reverse MR results showed that there was no causal relationship between the disease and the metabolites.Conclusion Urea has an inhibitory effect on cerebral infarc-tion,which may provide new ideas and new basis for the clinical treatment and scientific research of cerebral infarction in the future.关键词
