Abstract

To investigate the role of Salmonella Pullorum(ATCC19945)(WT,the same below)myceline-threonine protein kinase(YihE)in the pathogenic process,In this study,a gene deletion strain(ATCC19945ΔyihE)(KO,the same below)and a gene complementing strain(ATCC19945ΔyihE/pBR322-yihE)(RS,the same below)were constructed by λ-Red homologous recombination technology in the present study.Growth characteristics,anti-stress,locomotion,biofilm formation,drug resistance,anti-egg killing,bacterial adhesion,invasion and survival were determined for WT,KO and RS strains.The results showed that compared with the WT and RS strains,the KO strains had no significant difference in growth rate,locomotion and anti-egg killing while the anti-stress ability decreased by about 30%(except for heat stress of 42℃)the biofilm formation by about 40%.The formation ability of frillin as the main component of the biofilm,decreased while the visual appearance of cellulose as the main component of the biofilm did not change significantly.In addition,the sensitivity to quinolones was enhanced while adhesion,invasion and survival ability decreased.These results showed that the yihE gene was involved in multiple biological processes of Salmonella Pullorum and deletion of the yihE gene reduced relevant biological activities of Salmonella pullorum,which laid the foundation for further research on the pathogenic mechanism.
