DAPK2调控Ⅰ型干扰素启动子的初步研究
A Preliminary Study of DAPK2 Regulating the Promoter of Type Ⅰ Interferon

Abstract
To reveal the regulation of IFN-I by kinases in RLR signaling pathway,the dual luciferase reporter system and qRT-PCR were used in which a negatively regulated kinase DAPK2 was identified in this study.Moreover,DAPK2 was also studied for its regulatory specificity,kinase activity and regulation level.The results showed that DAPK2 specifically negative-regulated the production of type Ⅰ interferon in the RLR signaling pathway,which mainly concentrated on the IRF3-mediated pathway rather than the NF-κB-controlled inflammatory responses.After activating IFN-β promoters by signaling proteins at different levels,it was found that DAPK2 functioned between VISA and TBK1.The absence of DAPK2 kinase activity mutant in regulating RLR pathway suggested that the kinase activity was important in the regulation.This study provided a potential drug target and theoretical guidance for future application of DAPK2 in the treatment of zoonotic diseases caused by viruses.关键词
