Abstract

To establish a Kunming mouse model of porcine deltacoronavirus(PDCoV),8-week-old SPF female Kunming mice were inoculated with 107.6 TCID50/mL of PDCoV JS2211 strain in this study.The infected group was given 200 uL PDCoV by gavage needle and 100 μL PDCoV by intramuscular injection.The control group was given equal volume of normal saline in the same way as the infection group.The clinical signs of mice were observed every day.At 1,3,5,7,10,14 and 21 days post infection,3 mice in the infection group and 1 mouse in the control group were randomly sacrificed to collect organs for histopathological observation and virus distribution by qRT-PCR.The results showed that the infected mice developed intestinal inflating,edema,wall thinning,mesenteric bleeding and other gastrointestinal lesions on 7 d,14 d and 21 d post infection.The histopathological observations found intestinal villi were partially defected with low density,shortness,thinner walls.Additionally,obvious bleeding in the mesentery,superficial gastric mucosal epithelial cells were accompanied by a little erosion and ablative.Many inflammatory cells were seen in these tissues.qRT-PCR showed that a large number of viruses were detected in the intestine tissues of infected mice and the level of viruses peaked at 14 days post infection.In conclusion,this study provided a useful animal model for the evaluation of antiviral biological products and drugs for PDCoV infection.
