TGEV SHXB株组织毒对1月龄仔猪的致病性研究
Study on the Pathogenicity of TGEV SHXB Strain from Intestinal Contents in One-Month-Old Piglets

Abstract
Porcine Transmissible gastroenteritis virus(TGEV)causes severe vomiting,watery diarrhea,dehydration and death in suckling piglets.As the piglets grow up,the clinical signs become mild.However,whether TGEV is pathogenic to 1-month-old piglets has not been reported.This study was conducted to evaluate the pathogenicity of TGEV SHXB strain in 1-month-old piglets.Five 5-day-old piglets were divided into two groups.The first group was orally administered with 2 mL TGEV SHXB virus solution at titer of 105.0 TCID50/piglet,and the second group was orally administered with 2 mL DMEM.When the viral load peaked,the intestines were ground to inoculate next piglets.Subsequently,eight 1-month-old piglets were divided into two groups.The first group of 1-month-old piglets was orally administered with 10 mL TGEV SHXB tissues at titer of 106.50 copies/mL and the second group was orally administered with 10 mL DMEM.Anal swabs were daily collected post inoculation and the virus shedding was examined in fluorescence quantitative assay.Diarrhea of the infected piglets was observed and recorded.All piglets were terminated on 7 days post infection and the changes of intestinal tracts and other organs were examined.Duodenums,jejunums,ileums,cecums,colons and rectums were taken for pathological dissection and immunohistochemistry.The results showed that TGEV SHXB strain was highly pathogenic to 5-day-old piglets and their intestinal contents infected 1-month-old piglets.The infected piglets showed watery diarrhea on day 3 post infection and the viral loads peaked at about 108.0 copies/mL on day 4 post inoculation.Autopsy showed intestinal dilatation,intestinal wall thinning and congestion and mesenteric congestion.Histopathological examination showed severe atrophy and shedding of small intestinal villi accompanied by inflammatory cell infiltration.The results of immunohistochemistry showed that TGEV positive particles were detected in the epithelial cells of the duodenums,jejunums and ileums of the infected pigs.This study demonstrated for the first time that the intestinal contents of TGEV SHXB strain infected piglets had strong pathogenicity to 1-month-old piglets,which provided a new reference for the study of TGEV infection model in piglets.关键词
