Metformin Sensitizing the Inhibitory Effect of Gemcitabine on Human Pancreatic Cancer Cells and Its Mechanism

Abstract
Objective To investigate the mechanism of metformin in enhancing the sensitivity of the inhibitory effect of gemcitabine on human pancreatic cancer cells(aspc-1).Methods Cells were divided into the blank control group(group A,conventional culture medium),metformin group(group B,conventional culture medium with 5.00 mmol/L metformin),gemcitabine group(group C,conventional culture medium with 2.50 μg/L gemcitabine),and combined group(group D,conventional culture medium with 5.00 mmol/L metformin+2.50 μg/L gemcitabine).After treatment,cells in each group were cultured for 48 h.Cell apoptosis was detected by the flow cytometry,and the expression levels of P-glycoprotein(P-gp),hypoxia inducible factor-1α(HIF-1α),and phos phorylatod nuclear factor κB(p-NF-κB)proteins were detected by the Western blot.Results Compared with those in group A,the apoptosis rates of cells in groups C and D significantly increased(P<0.05).Compared with those in group C,the apoptosis rate of cells in group D significantly increased(P<0.05).Compared with those in group A,the expression levels of P-gp,HIF-1α,and p-NF-κB p65 proteins in group B significantly decreased(P<0.05),while the above indexes in group C significantly increased(P<0.05);compared with those in group C,the above indexes in group D significantly decreased(P<0.05).Conclusion Metformin can enhance the inhibitory effect of gemcitabine on human pancreatic cancer cell aspc-1,and its mechanism may be related to the down-regulation of P-gp to enhance the sensitivity of aspc-1 to gemcitabine.关键词
