Literature Analysis of Adverse Drug Reactions Induced by Olaparib
摘要
Abstract
Objective To summarize the occurrence pattern and characteristics of adverse drug reactions(ADRs)induced by olaparib,and to provide a reference for safe clinical drug use.Methods Case reports of olaparib-induced ADRs in the PubMed,Embase,Web of Science,CNKI,WanFang,and VIP databases were searched from the inception of each database to May 31,2024.Excel software was used for statistical analysis,and the systemic organ classification(SOC)of the ADR terminology in MedDRA 27.0 was used to classify ADRs.The occurrence characteristics,treatment methods,and prognosis of ADR were summarized.Results A total of 22 case reports of ADR induced by olaparib were included,involving 24 patients.Among the 24 patients,the age was in the range of 35-80 years old,with an average of 60.95 years old;there were four males(16.67%)and twenty females(83.33%);ADR occurred from 3 d to 13 months after medication,with approximately 58.33%of ADRs occurring within 3 months after medication;the systems/organs involved in ADRs included the respiratory system,chest and mediastinum,blood and lymphatic system,skin and subcutaneous tissue,liver and gallbladder,etc.In the treatment and outcome of 24 patients,20 cases improved after discontinuing medication,reducing dosage,using glucocorticoids,or symptomatic supportive treatment,one case deteriorated in general condition and died 5 months after medication,one case did not improve after treatment,two cases developed myelodysplastic syndrome,with one case undergoing bone marrow transplantation after discontinuing olaparib for 4 weeks without relief,and the other case died after 47 d of discontinuing olaparib.Conclusion Olaparib-induced ADRs can involve multiple systems/organs,and are more common in female patients.Drug monitoring should be strengthened during clinical use,and effective intervention should be given in time once ADR occurs to reduce the patient's medication risk.关键词
