Protective effect of exosomes derived from menstrual blood mesenchymal stem cells on cisplatin-injured human ovarian granu-losa cells KGN

Abstract
Objective To investigate the protective effect of menstrual blood mesenchymal stem cell-derived exosomes(MenSCs-EXOs)on cisplatin(DDP)-induced injury in human ovarian granulosa cell KGN and to explore the potential molecular mechanism.Methods MenSCs-EXOs were isolated by ultracentrifugation and identified by transmission electron microscope and Western blot.KGN cells were treated with different concentrations of DDP.Cell counting kit-8(CCK-8)assay was used to detect cell viability,and flow cytometry was used to detect cell apoptosis rate to screen out the optimal concentration of DDP.KGN cells were divided into three groups according to the treatment:control(Con)group(normal KGN medium),DDP group(medium containing the optimal DDP concentration),and DDP-EXOs group(medium containing the optimal DDP concentration and 10 μg/mL MenSCs-EXOs).After 48 hours of culture,CCK-8 assay was used to detect KGN cell viability,enzyme-linked immunosorbent assay was used to detect hormone secretion levels,flow cytometry was used to detect apoptosis,and Western blot was used to detect the expression levels of key regulatory proteins related to the transforming growth factor-β(TGF-β)/mothers against DPP homolog(SMAD)pathway.Results MenSCs-EXOs were successfully isolated by ultracentrifugation.A concentration of 3 mg/L DDP significantly inhibited KGN cell proliferation and induced apoptosis simultaneously(both P<0.05),so this concentration was selected as the optimal DDP concentration.Compared with the Con group,the DDP group showed decreased cell viability,increased apoptosis rate,reduced estradiol(E2)level,upregulated expressions of SMAD2,SMAD7,and SMURF2 proteins,and downregulated expressions of SMAD3 and SMAD4 proteins(all P<0.05).Compared with the DDP group,the DDP-EXOs group showed increased cell viability,decreased apoptosis rate,increased E2 level,downregulated expressions of SMAD2,SMAD7,and SMURF2 proteins,and upregulated expressions of SMAD3 and SMAD4 proteins(all P<0.05).Conclusion MenSCs-EXOs can effectively promote the proliferation and inhibit the apoptosis of DDP-injured granulosa cells,and their protective mechanism may be achieved by activating the TGF-β/SMAD signaling pathway.关键词
