PARP-1基因多态性与乳腺癌风险的相关性
Association between PARP-1 gene polymorphism with breast cancer susceptibility

Abstract
Objecitve:The aim of the present study was to examine whether single nucleotide polymorphisms(SNP)in the PARP-1 gene are associated with risk of breast cancer in the population of Gansu Province.Method:Four SNPs in the PARP-1 gene in a case-control study were genotyped,including 475 breast cancer cases and 944 healthy controls of Chinese Han woman in Gansu Province,China.Genotypes were determined by using the TaqMan® Array Card assays on the QuantStudioTM 12K Flex Real-Time PCR System.Results:We found that GA and AA genotype of rs1805408 had significant lower risk of breast cancer(adjusted odds ratio OR=0.372,95%CI 0.270-0.512,P<0.001;OR=0.584 95%CI 0.964-0.991,P<0.001)compared with GG genotype,and CT genotype of rs1805414 conferred significant reduced risk of breast cancer(adjusted odds ratio OR=0.626,95%CI 0.498-0.787,P<0.001)compared to CC genotype.Rs1805408 and rs1805414 also conferred a decreased risk of breast cancer in dominant models(AA+AG vs.GG,OR=0.465,95%CI 0.347-0.624,P=0.000;TT+CT vs.CC,OR=0.662,95%CI 0.543-0.821,P<0.001).Besides,Significant associations were also shown between the SNPs of PARP-1 and the status of p53,Ki67,ER and PR.Furthermore,it is also showed that cases carrying the PARP-1 haplotypes had significant associations of breast cancer compared with healthy controls.Conclusion:SNP variants in the PARP-1 gene exhibit a positive association with an elevated risk of breast cancer.These findings suggest that variants in PARP-1 gene may influence the risk of breast cancer.关键词
