外泌体滴鼻对抑郁小鼠行为的影响及单胺类递质释放水平的改变观察
Observation of the effects of exosome intranasal administration on the behavior of depressed mice and changes in monoamine neurotransmitter release levels

Abstract
Objective:To investigate the effects of intranasal administration of adipose-derived mesenchymal stem cell exo-somes(ADMSCs-EXO)on depressive behaviors in mice and its underlying mechanisms.Methods:Chronic binding stress was used to develop depression models,with EXO or PBS applied once daily for 8 weeks,and normal control mice were not treated.The depressive behavior was assessed using the suspended tail and forced swim experiment.Immunofluorescence was utilized to observe the brain entry pathway of EXO labeled with PKH26.Following additional behavioral testing,ELISA assay was conduct-ed to detect changes in monoamine neurotransmitters in the hippocampus.Results:After 72 h of intranasal administration of EXO,it reached brain regions such as the hippocampus.After 8 weeks of treatment,the improvement in depressive behavior in mice from the CMS+EXO group was statistically significant(P<0.05).Furthermore,statistically significant increases in the lev-els of 5-hydroxytryptamine,dopamine,and norepinephrine in the hippocampus were observed(P<0.01).Conclusion:Intranasal administration of ADMSCs-EXO can alleviate depressive behavior in mice and elevate monoamine neurotransmitter levels,sug-gesting that intranasal EXO may represent a novel therapeutic strategy for treating depression.关键词
