Abstract

Ganoderma sichuanense is a kind of macrofungi with high medicinal and economic values.For the extensive exploration and utilization of its natural resources,an analysis of internal transcribed spacer sequences(ITS)was conducted in this study on three wild Ganoderma strains collected respectively from Fujian and Zhejiang provinces in China,all of which were identified as belonging to the species of G.sichuanense.The above three wild strains of G.sichuanense(S-Ⅱ,S-Ⅲ,S-Ⅳ)together with one major domestic strain(S-Ⅰ)were used as research materials for biological characterization,cultivation trial,and detection of main ganoderic acids.Best growth conditions seemed to differ among individual strains of G.sichuanense with an optimal temperature of 30 ℃,an optimal carbon source of starch,an optimal nitrogen source of yeast extract,and an optimal pH value of 5.Cultivation experiments using artificial substitutes demonstrated that all of the three wild G.sichuanense strains were able to generate fruiting bodies and conduct sporulation in normal.Analysis of chemical substances within the fruiting bodies of tested strains by using ultra-performance liquid chromatography combined with quadruple-time-of-flight mass spectrometry(UPLC-QTOF-MS)led to the successful detection of 18 ganoderic acids that have already been reported to carry extensive pharmacological effects.It was very interesting to know from this study that the wild strain S-Ⅳrevealed its content of both 13 individual ganoderic acids or total ganoderic acids higher than that of cultivated strain.The above successful cultivation of three wild strains and discovery of one wild strain with a high content of ganoderic acids should provide available references for the future development of wild germplasm resources and breeding of new varieties of G.sichuanense.
