miR-147与miR-498在肺腺癌组织中的表达及与临床病理特征、生存预后的关系
Expression of miR-147 and miR-498 in Lung Adenocarcinoma and Their Relationship with Clinicopathological Characteristics and Survival Prognosis

Abstract
Objective To investigate the expression of microrNA-147(miR-147)and microrNA-498(miR-498)in lung adenocarcinoma tissues and their relationship with clinicopathological characteristics and survival prognosis.Methods A total of 103 patients with lung adenocarcinoma admitted from January 2021 to March 2023 were selected as the research subjects.The expression levels of miR-147 and miR-498 in cancer tissues and adjacent tissues were compared,and the expression levels of miR-147 and miR-498 in cancer tissues of patients with different clinicopathological characteristics were also compared.The relationship between miR-147,miR-498 and the clinicopathological characteristics of patients with lung adenocarcinoma was analyzed.The receiver operating characteristic(ROC)was used to analyze the diagnostic efficacy of miR-147 and miR-498 for lung adenocarcinoma,and miR-147 and miR-498 were classified as low-expression and high-expression based on the optimal cut-off value.The one-year survival rates of patients with different expression levels of miR-147 and miR-498 were compared.Results The expression levels of miR-147 and miR-498 in cancer tissues were lower than those in adjacent tissues(P<0.01).The expression levels of miR-147 and miR-498 in cancer tissues of patients with tumor diameter≥3 cm,TNM stageⅢ-Ⅳ,pleural invasion,lymph node metastasis,or poor differentiation were respectively lower than those in patients with tumor diameter<3 cm,TNM stageⅠ-Ⅱ,no pleural invasion,no lymph node metastasis,or moderate to high differentiation(P<0.01).Spearman correlation analysis showed that miR-147 and miR-498 were negatively correlated with the tumor diameter,TNM stage,pleural invasion and lymph node metastasis of lung adenocarcinoma,and positively correlated with the degree of differentiation(P<0.01).ROC curve analysis showed that the optimal cut-off value for miR-147 diagnosis was 1.46,and the area under the curve(AUC)was 0.753(95%CI:0.688,0.810).The optimal cut-off value for the diagnosis of miR-498 was 2.30,and the AUC was 0.765(95%CI:0.679,0.863).A total of 103 patients were followed up for one year,and 1 patient was lost to follow-up.The 1-year survival rate of the 102 patients who completed the follow-up was 84.31%(86/102).The 1-year survival rates of patients with low expression of miR-147 and miR-498 were 78.57%(55/70)and 77.61%(52/67),respectively,which were both lower than those of patients with high expression[96.88%(31/32)and 97.14%(34/35)],and the differences were statistically significant(P<0.05).Conclusion miR-147 and miR-498 are lowly expressed in lung adenocarcinoma,are significantly correlated with tumor diameter,TNM stage,pleural invasion,lymph node metastasis,and degree of differentiation,and have a significant impact on survival status.关键词
