摘要

Abstract

Objective To investigate the potential mechanism by which Guanxinkang(冠心康,GXK)regulates adipose tissue browning in ApoE-/-mice with atherosclerosis(AS)model by observing its effect on Trib1 in adipose tissue.Methods ApoE-/-mice with high-fat diet(HFD)were used to establish an AS model,which were then randomly divided into the model group,low/high-dose GXK groups,and atorvastatin group,while C57BL/6J mice were selected as the control group.After intervention,contents of total cholesterol(TC),triglycerides(TG),and low-density lipoprotein cholesterol(LDL-C)in serum of each group were measured.Oil Red O stai-ning was used to observe the pathological changes of aortic sinus.HE staining was used to observe the pathological changes of adipose tis-sue.Western blot(WB)was performed to detect the relative protein expressions of peroxisome proliferators-activated receptor gamma coactivator 1 alpha(PGC1α),mitochondrial brown fat uncoupling protein-1(UCP1),Tribbles homolog 1(Trib1)in adipose tissue.Results Compared with the control group,the model group showed increased serum levels of TC,TG and LDL-C(P<0.01),signifi-cantly enlarged aortic sinus plaques and adipocyte volume,disorganized cell arrangement,and decreased protein expressions of PGC1α,UCP1,and Trib1 in adipose tissue(P<0.01).Compared with the model group,the high-dose GXK and atorvastatin groups exhibited reduced levels of serum TC,TG,and LDL-C(P<0.05),decreased ratio of aortic sinus plaque to lumen area(P<0.01),smaller adipocytes with orderly arrangement,and increased protein relative expressions of PGC1α,UCP1,and Trib1 in adipose tissue(P<0.05).Conclusion GXK may ameliorate AS by promoting adipose tissue browning in ApoE-/-mice via mediating Trib1 in AS model.
