疏肝补肾方对卵巢早衰大鼠PI3K/AKT/mTOR信号通路的调节作用
Regulatory effect of Shugan Bushen Formula(疏肝补肾方)on the PI3K/AKT/mTOR signa-ling pathway of rats with premature ovarian failure

Abstract
Objective To elucidate the mechanism of action of Shugan Bushen Formula(疏肝补肾方,SGBSF)in treating premature o-varian failure(POF)rats through regulating the PI3 K/AKT/mTOR signaling pathway.Methods 40 SD rats were randomly assigned to the blank group(8 rats)and modeling group(32 rats)using the randomized digitaltable.The blank group was not intervened,while the modeling group was constructed using CUMS combined with cyclophosphamide chemical induction to establish a POF model.The model rats were further randomly divided into the model group,liver-soothing group,kidney-tonifying group,and liver-soothing and kidney-tonifying group.The liver-soothing group,kidney-tonifying group,and liver-soothing and kidney-tonifying group were gavaged with Chaihu Shugan Powder(柴胡疏肝,CHSGP),Zuogui Decoction(左归饮,ZGY),and SGBSF,respectively.The blank group and the model group were gavaged with normal saline for 4 consecutive weeks.The weight and behavioral changes of rats in each group were observed,serum levels of FSH,E2,AMH,and INHB were detected by ELISA,pathological changes in the ovarian tissues were ob-served by HE staining,and protein expressions of PI3K,AKT,and mTOR in the ovarian tissues were detect by Western blot(WB)and immunohistochemistry(IHC).Results The estrous cycle of the model group was disrupted,and the consumption of sugar water and total exercise distance were significantly reduced compared to the blank group.The serum FSH level was significantly increased,while the E2 level was significantly reduced(P＜0.01),indicating successful modeling.The liver-soothing and kidney-tonifying group showed a decrease in the number of blocked follicles in the ovarian tissues compared to the model group,with more follicles at all levels visible.The sugar water consumption and total exercise distance were both significantly increased;the FSH level was significantly reduced,while E2,AMH,and INHB levels were significantly increased.The expressions of PI3 K,AKT,and mTOR proteins in the ovarian tissues were significantly increased(P＜0.05 or P＜0.01),and the therapeutic effect of liver-soothing and kidney-tonifying group was better than those of the liver-soothing group and kidney-tonifying group.Conclusion SGBSF may have a therapeutic effect on POF rats by regula-ting key proteins in the PI3K/AKT/mTOR signaling pathway.关键词
